Dolinnov develops a first-in-class non-opioid therapy for chronic pain, one of the largest unmet medical needs worldwide. Despite recent progress, current approaches targeting single ion channels (Nav1.7, Nav1.8) face key limitations due to redundancy, as other channels can compensate over time, limiting long-term efficacy.

Dolinnov addresses this challenge by targeting a novel and highly differentiated peripheral mechanism. Instead of blocking ion channels, Dolinnov modulates a key regulator of neuronal excitability using lipid-conjugated antisense oligonucleotide (L-ASO) technology to selectively target the dorsal root ganglion (DRG), where chronic pain originates.

This approach enables the reduction of pathological nerve activity while preserving normal sensory function. Dolinnov’s lead candidate, DOL-001, has demonstrated strong efficacy across multiple in vivo pain models, with a long-lasting effect and robust translational potential supported by human DRG expression and mechanistic data.

By targeting ion pumps rather than ion channels, Dolinnov offers a precise, peripheral, non-opioid alternative that avoids central nervous system toxicity and overcomes the limitations of current analgesics.
